Anti goat IGF binding protein 3 antibodies from chicken egg yolk

The use of chickens for the production of highly specific antibodies to mammalian proteins offers a number of advantages over more conventional methods. Antibody responses to IGFBP-3 immunogen were apparent in the serum of chickens after only two inoculations with low levels of the immunogen. These specific antibodies subsequently appeared in the chicken egg yolk at levels of approximately 40% of those estimated in serum.

A simple, rapid and reproducible two step method for the extraction and purification of the immunoglobulin fraction from egg yolk using octanoic acid is described. Relatively large quantities of specific antibodies can be obtained by these procedures. Egg yolk antibodies generated in this study will be used to develop an ELISA for measuring levels of IGFBP-3 in plasma and other body fluids. This approach is designed to overcome some of the difficulties associated with the use of ligand blotting analysis to quantify IGFBPs.
